Impact of three anticoagulants and their storage time on hematological parameters of snow trout, Schizothorax labiatus, habiting in river Sindh of Indian Himalayan region

Abstract

Blood analysis is a critical and potent diagnostic tool used in the field of fisheries to determine the well-being status of the fish. However, internal and external influences, such as anticoagulants and storage time, can have a significant impact on fish hematological values resulting in species-specific reactions. Because of the short clotting periods, fish blood generally needs the inclusion of an anticoagulant agent; therefore, choosing the right anticoagulant is imperative for achieving accurate blood test results. The aim of current study is to analyze the impact of three different anticoagulants (lithium heparin, di-potassium EDTA, and sodium citrate) on various hematological characteristics of snow trout, Schizothorax labiatus, and their effect on different intervals of storage time at + 4 °C. The results revealed that Hb, Hct, MCH, and MCV in all the samples treated with K2EDTA produced significantly (P < 0.05) higher values than other two anticoagulants. While, total RBC count was recorded significantly (P < 0.05) higher in lithium heparin–treated samples. However, total WBC count and MCHC in all the three anticoagulant-treated samples exhibited insignificant (P > 0.05) difference. After analysis at 0 h, the blood samples were refrigerated at + 4 °C for 24 h, 48 h, and 72 h, respectively. Statistical evaluation revealed significant (P < 0.05) impact of storage time on values of Hb, RBC, Hct, MCH, MCHC, and MCV except WBC after 24 h of time interval. Hence, on the basis of above mentioned findings, it is suggested that for fish blood analysis, lithium heparin provides more reliable data with a higher level of anticoagulant dependability in S. labiatus and also the blood samples must be analyzed within 24 h after collection when stored at + 4 °C as indicated in the present study that longer storage affects the hematological profile of the fish.
