Astrocytoma Cell Line: Role of Brain Natriuretic Peptide

Abstract

Brain natriuretic peptides (BNP) is widely distributed in central nervous system (CNS) structures like C type NP (CNP) and more than atrial NP (ANP). In contrast, NP receptors (NPRs) are mainly localized in astroglial cells in several brain regions, and astrocytes express predominantly the NPR-A subtype. Thus, NPs play an important role in the regulation of astrocyte functions including anti-apoptosis, anti-proliferation, and neuron-glia communication. The human astrocytoma cell line U373MG releases BNP, and hypoxia enhances the gene expression and release of BNP. Hypoxia-induced apoptosis in U373MG is increased by knockdown of BNP, whereas it is dampened by exogenous BNP, suggesting that not only endogenous but exogenous BNP functions as an anti-apoptotic molecule. BNP inhibits DNA synthesis by binding to NPR-C and localizing to nucleus of cancer cells, and exerts anti-proliferative effects in human glioblastomas cell line. Since NPs circulate normally in the human body and have no side effect on normal cells, BNP may be a novel therapeutic molecule against brain tumor.
