Homogeneous Glycoproteins: Overview

Abstract

The heterogeneous nature of glycoproteins makes it difficult to understand how glycosylation affects the structure and function of glycoproteins. Development of new methods to prepare homogeneous glycoproteins with well-defined glycan structures is thus important to tackle this issue. This chapter describes various methods available for the synthesis of homogeneous glycoproteins for use to study the effect of glycan in glycoprotein folding, stabilization, and function, and to develop single glycoforms with optimal activity as pharmaceuticals. A major problem in the field is the lack of methods for modification of a specific glycan and a specific monosaccharide on a glycoprotein with multiple glycans, and this unsolved problem remains to be a major challenge in glycoscience.
