Zusammenfassung

Die markerfreie, quantitative In-vitro-Bildgebung lebender Zellkulturen mit Lichtmikroskopie ist ein wichtiges Werkzeug für verschiedenste Forschungsfelder in den Biowissenschaften. Die digitalholografische Mikroskopie (DHM) ermöglicht die Charakterisierung von Zellen und Geweben und die Quantifizierung zellulärer Prozesse ohne Färbung oder Markierung. Das Verfahren ist schnell und nicht invasiv, biologische Proben können daher über einen längeren Zeitraum vermessen und anschließend mit anderen Verfahren weiter untersucht werden. Das Kapitel befasst sich zunächst mit einer kurzen Einführung in die DHM zur Analyse von lebenden Zellen und Gewebeschnitten. Des Weiteren werden Verfahren zur Extraktion biophysikalischer Parameter aus quantitativen DHM-Phasenkontrastbildern erläutert. Schließlich wird anhand ausgewählter Anwendungen der Einsatz in der In-vitro Lebendzellanalyse und bei der Quantifizierung von für immunologisch relevanten physikalischen Gewebeeigenschaften illustriert.
