Zusammenfassung

Das Immunsystem ist die effektivste Waffe des Körpers, um Krankheitserreger, aber auch Krebszellen abzuwehren. Trotzdem entkommen manche Tumorzellen dieser Immunüberwachung. Grundlegende Voraussetzungen für das Verständnis von zielgerichteten Strategien zur Beeinflussung des Immunsystems bei der Bekämpfung von Krebs sind, die Prinzipien der Entstehung, Aktivierung und Regulation von Immunzellen sowie die molekularen Strukturen und Interaktionen zu kennen. In diesem Kapitel werden daher zunächst die wesentlichen Mechanismen vorgestellt, mit denen Immunzellen des angeborenen und adaptiven Immunsystems ihre Abwehrfunktion ausüben. Weiterhin werden bisher identifizierte Immune-Escape-Strategien von Tumorzellen beschrieben, die zu einer Tumorentwicklung und -progression führen können. Auf der Basis dieser grundlegenden immunologischen Erkenntnisse konnten in den vergangenen Jahren mehrere sehr erfolgreiche immunonkologische Therapiestrategien entwickelt und in die klinische Anwendung gebracht werden.
