Abstract
A L-valine production strain Brevibacterium flavum C1(Ile−) was mutagenized using UV-irradiation and diethyl sulfate and was screened on the selective plates. The selective plates contained 4 g/L 2-thiazole-DL-alanine (2-TA) or 5 g/L α-aminobutyric acid (α-AB). After mutagenesis and selection, 1500 mutants were obtained, and 57 were found to be resistant to 2-thiazole-DL-alanine and α-aminobutyric acid. Following determination of the L-valine produced by these 57 mutants in fermentation medium in shaken cultures, a mutant CC5 (Ile−, 2-TAr, α-ABr) showed highest L-valine production ability than the original strain.
