Abstract
If an unbalance of oxidants and antioxidants appears in the skin, oxidative stress is the consequence, and an excess of reactive oxygen species can cause severe cell damage associated with skin carcinogenesis and accelerated skin aging. In this chapter, it is shown that electron paramagnetic resonance spectroscopy is able to measure the antioxidant status of the skin by investigating the radical scavenging activity of applied test radicals. The direct reaction of cutaneous antioxidants can be studied using the application of the test radical TEMPO (2,2,6,6-tetramethyl-1-piperidinyloxy), which is exponentially reduced. These reduction kinetics can be exponentially fitted and the resulting rate constant is a measure of the antioxidant status. Irradiation induced short-living radicals can be investigated by their competitive reaction between the applied test radical PCA (3-carboxy-2,2,5,5-tetramethyl-1-pyrrolidinyloxy) and the cutaneous antioxidants. It could be shown that orally and topically supplemented antioxidants increase the cutaneous antioxidant status. Systemic antioxidants applied in a physiological concentration can increase the antioxidant status by 50 %. Furthermore, compounds which increase the rate constant also provide protection against stress induced radicals as formed during VIS/NIR irradiation. Topical and oral application can be combined providing an optimum of protection against premature skin aging and development of cancer.
