Abstract
One of the important factors influencing the separation efficiency and sensitivity of the capillary electrophoresis microchip is the background material. In this work a microchip capillary electrophoresis (MCE) featuring Agarose gel as a separating medium was fabricated and tested in the laboratory. The design was based on polydimethysiloxane (PDMS) engraved microchannel with platinum electrodes etched onto glass substrate. Operated under relatively low electric field of 24 V/cm and sample volume of 1.5 μl, the new device was able to separate DNA fragments with resolution consistently higher than 100 nA. Separation is achieved in less than 45 minutes.
