Abstract

Coenzyme Q10 (CoQ10) is an essential part of the mitochondrial respiratory chain. Here, we describe an accurate and sensitive liquid chromatography tandem mass spectrometry (LC-MS/MS) method for determination of mitochondrial CoQ10 in isolated mitochondria. In the assay, mitochondrial suspensions are spiked with CoQ10-[2H6] internal standard, extracted with organic solvents, and CoQ10 quantified by LC-MS/MS using multiple reaction monitoring (MRM).
