Abstract

Piperazic acid and its congeners are constituents of many structurally complex natural products. Their conformational rigidity and other advantageous physicochemical properties have motivated the incorporation of these non-proteinogenic amino acids into pharmaceutical agents and drug leads. This review provides an overview of recently published research on piperazic acids, their origins, chemical syntheses, and applications in constrained, biologically active peptides and peptidomimetics.
