Riassunto
L’attività biologica o tossicologica di un composto chimico può essere studiata servendosi di diversi descrittori molecolari. Fra questi rivestono una grande importanza le proprietà chimico-fisiche quali acidità, lipofilia e solubilità. Queste tre proprietà sono di facile interpretazione e risultano molto utili per la spiegazione di complessi fenomeni biologici in termini meccanicistici. La loro determinazione sperimentale richiede spesso tempi lunghi ed è quindi utile poter disporre di strumenti di calcolo in silico veloci e robusti in grado di stimare il valore di queste proprietà anche per composti non ancora sintetizzati. In questo capitolo saranno descritte alcune delle metodologie più utilizzate per la predizione in silico di queste tre proprietà. Una particolare attenzione sarà rivolta ai metodi che, grazie all’utilizzo di nuovi dati sperimentali, permettono l’addestramento dei predittori al fine di migliorarne l’accuratezza.
