Abstract

Mammalian alpha1,6-fucosyltransferase (FUT 8) catalyses the transfer of a fucose residue from a donor substrate guanosine 5′-diphosphate-beta-l-fucose to the reducing terminal N-acetylglucosamine (GlcNAc) of the core structure of an asparagine-linked oligosaccharide. The enzymatic product is called as core-fucosylated N-glycans, which are ubiquitously distributed in many mammalian glycoproteins. Serum levels of fucosylated alpha fetoprotein are very low under normal conditions, but it increases dramatically in malignant diseases and therefore it is one of the best biomarkers for primary hepatomas (see reviews by Miyoshi et al.). Core fucosylation plays a pivotal role in ADCC activity in antibody therapy (see reviews by Sato et al.). KO mice of Fut 8 exhibit severe growth retardation and develop lung emphysema. Crystal structure of this enzyme was reported very recently by our group.
