Abstract
Leptin is an adipose tissue-derived hormone implicated in the pathogenesis of atherosclerosis. Peripheral blood monocytes were incubated for 7 days to induce differentiation into macrophages. Under these conditions, exogenous leptin was added to examine its effect on macrophage foam cell formation. Leptin (5 nmol/L) significantly increased acetylated LDL-induced cholesteryl ester accumulation. Leptin increased both acyl-coenzyme A: cholesterol acyltransferase (ACAT) activity and ACAT-1 protein expression. Among the four ACAT-1 mRNA transcripts, two shorter transcripts (2.8- and 3.6-kb) were up-regulated upon leptin treatment. Importantly, leptin receptor expression was up-regulated during monocytic differentiation into macrophages, indicating stage-specific leptin signaling in human monocyte-macrophages.
