Abstract

The JAK-STAT pathway, originally identified in the interferon signaling pathway, has been shown to play an essential role in cytokine-mediated biological responses mainly through the analysis of knockout mice. The JAK family consists of four members (Jak1-3 and Tyk2), and the STAT family consists of seven members (Stat1–4, Stat5a, Stat5b, and Stat6). The JAK family of proteins plays an essential role mediated by cytokine receptors, whereas STAT proteins are responsible for their specific cytokines. In addition, there is a considerable body of evidence demonstrating that activity of the JAK-STAT pathway is regulated by several mechanisms, including the sacs family of proteins induced by activation of the JAK-STAT pathway. Gene targeting of the SOCS family revealed that individual sacs proteins negatively regulate some cytokine-mediated signaling pathways.
