Multiplexed Immunoassays for Protein Profiling in Human Body Fluids

Abstract

This chapter describes the development and use of suspension antibody microarrays for protein profiling in several human body fluids. In suspension microarrays, which allow the simultaneous analysis of a variety of analytes within a single experiment, capture antibodies are coupled onto color-coded microspheres. Applications are described, with emphasis on analyses of proteins present in different types of body fluids, like serum or plasma, tears, cerebrospinal, pleural, and synovial fluids, and cell culture supernatants. The chapter is divided into the generation of suspension microarrays, sample preparation, processing, and validation of analytical performance.
