Zusammenfassung

Bei T-Zell-vermittelten Autoimmunerkrankungen wie der rheumatoiden Arthritis (RA) werden Th1-Zellen und deren Zytokine, insbesondere Interferon-γ (IFN-γ), für die Induktion und Unterhaltung der chronischen Entzündung und Gewebsschädigung verantwortlich gemacht. In Untersuchungen an experimentellen Modellen konnte aber inzwischen gezeigt werden, dass IFN-γ durchaus auch antiinflammatorisch wirken kann. In diesem Beitrag werden jüngste Ergebnisse diskutiert, die zeigen, dass IFN-γ – neben seinen bekannten proinflammatorischen Effekten – ein ganz wichtiger Regulator von Entzündungs- und Immunreaktionen ist. Solche selbstregulatorischen Prozesse scheinen enorm wichtig zu sein, um überschießende Reaktionen zu hemmen und die Homöostase des Immunsystems zu erhalten oder wieder herzustellen.

Abstract

In T cell-mediated autoimmune diseases such as rheumatoid arthritis, Th1 cells and their cytokines, especially interferon-γ (IFN-γ), are responsible for the induction and persistence of chronic inflammation and tissue destruction. But emerging evidence from experimental models has demonstrated that IFN-γ also possesses unexpected anti inflammatory properties. The recent data discussed in this article indicate that beside the well-known proinflammatory efficacy, IFN-γ may function as a master regulator of inflammation and immune responses. Such self-regulatory processes seem to play an important role in the inhibition of excessive responses and to maintain or reestablish homeostasis of the immune system.
