Comparison of dentin formed in vivo and dentin mineralized in vitro by contact microradiography: Effect of mg in mineralizing solution

Abstract

Dentins of rats were demineralized and the sections mineralized in vitro. Whether the distribution of mineral of the sections mineralized in vitro reproduces it of dentins formed in vivo and whether the distribution correlates with the nature of the matrices of dentins were examined by contact microradiography (CMR) and staining with hematoxylin. The abnormal layers induced to the dentins of rats administered Ni-salt or Mn-salt were chosen as the subjects of the examination.

The results showed the abnormal layers of the sections mineralized in the mineralizing solutions containing Mg were similar to the abnormal layers formed in vivo, but the abnormal layers of the sections from the mineralizing solutions containing no Mg were quite different from those in vivo. This suggested that Mg possibly also plays an important role in mineralization in vivo.

The abnormal layer induced by Ni-salt was strongly stained with hematoxylin in the demineralized sections of fixed dentins but not stained in unfixed dentins. In both fixed and unfixed dentins, the abnormal layers of the sections mineralized in vitro showed the same feature on the CMR, suggesting that substance strongly stained with hematoxylin is not a direct cause in the formation of the abnormal layers.
