Mikrochemischer Nachweis von o-Dioxo- und Oxomethylenverbindungen

Zusammenfassung

Die Reaktion der 1,2-Dioxime mit Nickelsalzen, deren Verwendung zum Nickelnachweis allgemein bekannt ist, soll in der vorliegenden Arbeit zum Nachweis von 1,2-Dioxoverbindungen durch Oximierung und anschließende Prüfung mit Nickelsalz herangezogen werden. Die Versuche zeigen, daß dieser Nachweis gelingt, abgesehen von jenen Ausnahmsfällen, wo es sich um Dioxoverbindungen von chinoidem Charakter handelt, wie Phenanthrenchinon usw., auf die das Hydroxylamin reduzierend einwirkt, so daß die Oximbildung ausbleibt.

Der Nachweis solcher o-Diketone gelingt dagegen mit 5-Dimethylamino-2-aminophenol durch Bildung eines intensiv blauen Oxazinfarbstoffes; diese letztere Reaktion versagt andererseits gerade bei jenen (z. B. aliphatischen) Dioxoverbindungen, die zur Oximbildung geeignet sind, so daß diese beiden Unterklassen gut unterschieden werden können.

Schließlich wird erwähnt, daß Oxomethylenverbindungen durch selenige Säure in ziemlich spezifischer Weise zu o-Dicarbonylverbindungen oxydiert werden, die sich anschließend nach Oximierung durch die Nickelsalzreaktion nachweisen lassen; so kann die letztere zum indirekten Nachweis der Gruppe —CO—CH2— verwendet werden, wenn man sich vergewissert hat, daß keine der — in der Arbeit diskutierten — anderen Gruppierungen vorhanden ist, die bei der Oxydation mit seleniger Säure ebenfalls zwei Carbonyle in o-Stellung liefern würden.

Summary

The reaction which 1,2-dioximes give with nickel salts has been used, as is generally known, for the detection of nickel. It is intended, in the paper present, to apply that reaction for the detection of 1,2-dioxo compounds. The test is carried out by converting the unknown substance to an oxime which, in its turn, is identified by addition of a nickel salt. It is shown by experiments that the test described can be carried out with success except in such unusual cases as in the presence of dioxo compounds of chinoid character, like phenanthrene quinone, etc. Those substances are reduced by the hydroxylamine and, consequently, oxime fails to appear.

In the contrary, such o-diketones can be successfully detected by means of 5-dimethylamino-2-aminophenol. Hereby, an oxazine dyestuff of intensive blue colour is formed; on the other hand, this latter reaction fails to appear with those very dioxo compounds (e. g., aliphatic ones) which are able to form oximes. Thus, both of those sub-classes can be well distinguished from each other.

It is mentioned, at last, that oxo methylene compounds can be oxidized, in a rather specific way, by means of selenious acid. As a result, o-dicarbonyl compounds are formed; these are converted to the respective oximes which, subsequently, can be detected by the nickel salt reaction. Accordingly, that reaction can be used for an indirect detection of the —CO—CH2— group. It has to be ascertained beforehand, that none of the other groupings — discussed in the paper in hand — is present, which also would yield two carbonyls in ortho position by being oxidized with selenious acid.

Résumé

La réaction des 1,2-dioximes avec des sels de nickel, dont l'emploi pour le dosage du nickel est bien connue, sera employée dans ce travail pour le dosage des combinaisons 1,2-dioxo par la préparation de l'oxime et indication suivante avec le sel de nickel. Les expériences montrent, que ce dosage est possible, abstraction faite des cas où il s'agit des combinaisons dioxos d'un caractère chinoïde, comme p. ex. phenanthrenchinone, qui sont reductées par l'hydroxylamine, de sorte que la formation d'un oxime n'a pas lieu.

Le dosage de telles o-dicétones réussit cependant avec 5-dimethylamino-2-aminophenol par la formation d'une matière colorante d'oxazine intensive bleue; cette réaction-ci ne fonctionne justement pas chez de telles combinaisons, p. ex. aliphatiques, qui sont capable de la formation d'un oxime, de sorte que ces deux classes peuvent être bien differenciées.

Enfin on mentionne, que les combinaisons oxymethylènes s'oxydent assez spécifiquement par acide sélénieux à des combinaisons o-dicarbonyles, qui peuvent être dosées par préparation de l'oxime et réaction du sel de nickel; cette réaction-ci peut être employée comme dosage indirecte du groupe: —CO—CH2—, si l'on s'est assuré de l'inexistence d'autres groupes — discutés dans ce travail-ci — qui fournissaient aussi deux o-carbonyles par l'oxydation avec acide sélénieux.
