Composition and seasonal variations of lipid components in the crayfish, Orconectes limosus

Summary

  1. 1.

    Lipid components of the crayfish, Orconectes limosus, were analyzed by thin-layer chromatography with respect to sex and seasonal variations. Up to 17 lipid classes could be identified and quantitatively determined in the following organs: abdominal muscle, hepatopancreas, testicles, ovary, gills, integumental tissue, nervous tissue, antennal gland, heart, stomach, gut, hemolymph, carapax, “rest body”, and exuvie. The different lipid classes were as follows: monoglycerides, α, α′-diglycerides, α, β-diglycerides, triglycerides, sterols, sterol esters, free fatty acids, fatty acid methylesters, carotenoids, hydrocarbons, phosphatidic acids, phosphatidyl choline, lysophosphatidyl choline, phosphatidyl ethanolamine, phosphatidyl serine, phosphatidyl inositol, and sphingomyeline.

    The total lipid content of a crayfish ranges between 5 and 17 mg/g fw. and varies greatly between organs. Higher values were found in the total body and most organs of the female compared with those in the male. The same could be realized for the triglycerides while the hydrocarbon levels showed an opposite tendency. Contrarily the distribution of phospholipids is highly uniform. In all organs the most important fraction is phosphatidyl choline accounting for 50–60% of total phospholipids, followed by the phosphatidyl ethanolamine fraction with about 25%.

    An annual cycle in the lipid content could be stated. The monthly analysis of total lipids allows to distinguish four cyclic periods. During the winter months the lipids decline to a minimum between May and June. A sharp rise follows with its maximum immediately before moult. The lipids being consumed during the moult reach a second minimum at the end of August. The cycle begins again with a storage of lipids leading to a winter maximum in October (cf. Fig. 3). Phospholipids and neutral lipids contribute to the lipid change of the different organs in a various extent. Organs with most excessive changes are hepatopancreas, integumental tissue, and abdominal muscle. During the period from October to March the ovary accumulates lipids obviously concerned with the process of egg maturation. It can be supposed a transport of neutral lipids from the hepatopancreas to the ovary. The total lipid content of the male hepatopancreas is constant in this time. For both sexes equally the hepatopancreas show the moulting and winter maxima.

    An Männchen und Weibchen des Flußkrebses Orconectes limosus wurde das Lipidspektrum zu mehreren Jahreszeiten dünnschichtchromatographisch analysiert. In den Organen: Schwanzmuskel, Mitteldarmdrüse, Hoden, Ovar, Kiemen, Integumentgewebe, Bauchmark, Antennendr, use, Herz, Magen, Enddarm, Hämolymphe, Carapax, Restkörper und in der Exuvie konnten bis zu 17 Lipidklassen identifiziert und quantitativ bestimmt werden. Es sind dies: Monoglyceride, α, α′-Diglyceride, α, β-Diglyceride, Triglyceride, Sterine, Sterinester, freie Fettsäuren, Fettsäuremethylester, Carotinoide, Kohlenwasserstoffe, Phosphatidsäuren, Phosphatidylcholin, Lysophosphatidylcholin, Phosphatidyläthanolamin, Phosphatidylserin, Phosphatidylinosit und Sphingomyelin.

    Der Gesamtlipidgehalt der Tiere liegt zwischen 5 und 17 mg/g Frischgewicht. Er variiert sehr stark innerhalb der Organe und ist bei den ♀ im gesamten Krebs und in den meisten Organen generell höher als bei den ♂. Das gleiche gilt für die Triglyceride und umgekehrt für die Kohlenwasserstoffe. Die Phosphatide zeigen ein weitaus einheitlicheres Muster. Die größte Fraktion ist in allen Organen Phosphatidylcholin (mit etwa 50–60% der Phosphatide); die zweitgrößte in den meisten Organen Phosphatidyläthanolamin (mit etwa 25% der Phosphatide).

    Der Lipidgehalt der Krebse verändert sich in einem Jahreszyklus. Bei der Bestimmung der Gesamtlipide in monatlichen Abständen lassen sich vier Perioden unterscheiden. In den Wintermonaten sinken die Lipide bis zu einem Minimum zwischen Mai und Juni. Danach folgt ein steiler Anstieg, der kurz vor der Häutung seinen Höhepunkt erreicht hat. Die Lipide, die während der Häutung verbraucht werden, erreichen schon Ende August wieder ein Minimum. Der Zyklus beginnt erneut mit einer Speicherung von Lipiden ab September und führt zum Herbstmaximum im Oktober (vergl. Abb. 3). In den Organen sind Phosphatide und Neutralfette auf sehr verschiedene Weise an den Veränderungen der Gesamtlipide beteiligt. Den größten Anteil daran haben Mitteldarmdrüse, Integumentgewebe und Schwanzmuskel. Im Zeitraum Oktober bis März speichert das Ovar im Zusammenhang mit der Eireifung Lipide, deren Neutralfettanteil offenbar aus der Mitteldarmdrüse stammt. Der Gesamtlipidgehalt der Mitteldarmdrüse des ♂ bleibt in dieser Zeit konstant. An den Häutungs- und Herbstmaxima haben die Mitteldarmdrüsen beider Geschlechter gleichermaßen Anteil.
