Summary
Twenty-seven spontaneous mutants were selected for streptomycin (50 μg/ml) resistance from Bacillus stearothermophilus strain 799. All of them were derived from independent mutational origins. Of these, ten mutants (about 35%) were found to be resistant to streptomycin, thirteen (about 50%) were fully and four (about 15%) were partially dependent on streptomycin for growth. The last mutants grow poorly without streptomycin but grow well with it. One of the ten resistant mutants isolated was partially sensitive to higher concentrations of streptomycin, while all the rest showed good growth with up to 500 μg/ml streptomycin. From some of the dependent mutants, spontaneous revertants which have lost the property of streptomycin dependence were isolated. 30 S ribosomal proteins from these five classes of mutants were analyzed by two-dimensional gel electrophoresis. Thus S12 was found to be affected in some of the resistant and fully dependent mutants, while S4 or S5 were altered in some of the independent revertants. One of the mutants partially dependent on streptomycin were found to have an alteration in S5. Another mutant partially sensitive to higher concentrations of streptomycin appears to have an alteration also in S5. The last two mutants are new classes of streptomycin mutants.
