Abstract
Britten and Kohne (1) have shown that the presence of repeated DNA sequences in eukaryotic organisms interferes with the true estimates of DNA·DNA and DNA·RNA reactions in annealing experiments. Neurospora crassa DNA has been reported to contain repeated DNA sequences (2). This might account for why no more than 20% DNA·DNA (3) and 9% DNA·RNA (Mahadevan and Bhagwat, this volume, Chapter 18) reactions were obtained using unfractionated DNA and RNA of the same N. crassa mycelial cells. A true estimate of the transcription of genetic information from DNA into complementary RNA in eukaryotic cells is needed for a better understanding of the control of cell and tissue differentiation. Using the conditions described in this chapter, it has been possible to arrive at an almost complete (98%) DNA·DNA and about 30% (where the theoretically maximum possible estimate is 50%) DNA·RNA hybridization in N. crassa at the exponential growth phase.
